ABSTRACT OF THE DISCLOSURE Methods for the production of steroids (sapogenins) and steroidal glycosides (saponins) from cell, root-cell and shoot tissue cultures of the Yucca plant. The cultures were established and maintained in a suitable nutrient plant cell medium supplemented with growth regulators. The desired products are extracted from the cultures using conventional extraction techniques. Greater total production of steroidal glycosides occurred in root-cell, cell, and shoot tissue cultures in that order. Although the cultures may be grown under conditions ranging from total darkness to total light, steroidal glycoside production was highest in root-cell cultures grown continuously in the light as compared to those grown in the dark. Root-cell culture senescence also enhanced the yield of total steroidal glycosides. Steroids are obtained from the steroidal glycosides by acid hydrolysis to remove the sugar moieties. The production of radio- labeled steroids and steroidal glycosides is disclosed. The steroidal glycosides produced by the method of the pre- sent invention have varied known uses. Thus, for example, as disclosed in Rothman U.S. Patent No. 2,715,122 steroidal saponin, which is a steroidal glycoside, may be useful in the synthesis of cortisone and other physiologically active substances. McKeen U.S. Patent No. 3,144,377discloses that sapongenins, which are steroids, are growth promoters useful in animal feeds and that the corresponding glycosides may be similarly used. Further, in the Merck Index Tenth Edition 1983 at No. 8218 there is shown that the steroidal glycosides may be used to form oil-in-water emulsions and to serve as pro- tective colloids, and also shows that the steroids may be used in the manufacture of steroidal hormones and compounds of the pregnane series. The use in animal feeds is of great commercial interest at the present time.
