A promoter-transactivator system achieves regulated high-level gene expression in proliferation-controlled mammalian cells. The novel composite promoter contains constitutive enhancer elements that allows a basal expression level as high as the levels achieved with a very efficient viral promoter. In addition, the promoter encodes sequences that are bound by a transactivator whose activity can be regulated. By the simple addition of medium supplements expression levels can be achieved above those from the conventional promoter.

Selon cette invention, un système de promoteur-transactivateur effectue l'expression régulée de gènes de haut niveau dans des cellules mammaliennes contrôlées par la prolifération. Le nouveau promoteur composite contient des éléments constitutifs renforçateurs qui permettent d'atteindre un niveau d'expression basale aussi élevé que ceux atteints avec un promoteur viral très efficace. En outre, le promoteur code des séquences qui sont liées par un transactivateur dont l'activité peut être régulée. En additionnant simplement des suppléments du milieu on peut atteindre des niveaux d'expression supérieurs à ceux du promoteur conventionnel.
