The present invention is concerned with a variant blood coagulation component, which is substantially homologous in amino acid sequence to a wild-type blood coagulation component capable of exhibiting anticoagulant activity in the protein C-anticoagulant system of blood and selected from protein C (PC) and activated protein C (APC), said variant component being capable of exhibiting an anticoagulant activity, that is enhanced in comparison with the anticoagulant activity expressed by the corresponding wild-type blood coagulation component, and said variant component differing from the respective wild-type component, in that it contains in comparison with said wild-type component at least one amino acid residue modification in its N- terminal amino acid residue sequence that constitutes the Gla-domain of protein C. The present invention is also concerned with methods to produce such variants based on DNA technology; with DNA segments intended for use in the said methods; and with use of said variants for therapeutic and diagnostic purposes.
La présente invention concerne une variante de composante de coagulation sanguine qui a une séquence d'acides aminés sensiblement homologue à celle d'une composante de coagulation sanguine de type sauvage capable d'avoir une activité d'anti-coagulation dans le système d'anti-coagulation à protéine C du sang et est choisie entre la protéine C (PC) et la protéine C activée (APC). Selon l'invention, ladite variante de la composante est capable d'avoir une activité d'anti-coagulation supérieure à l'activité d'anti-coagulation exprimée par la composante de coagulation sanguine de type sauvage, et ladite variante de la composante diffère de la composante de type sauvage respective en ce qu'elle contient, en comparaison avec ladite composante de type sauvage, au moins une modification de fragment d'acides aminés dans sa séquence de fragment d'acides aminés de terminal N qui constitue le domaine Gla de la protéine C. Cette invention concerne également des procédés permettant la production de variantes de ce type à partir de la technologie d'ADN, des segments d'ADN destinés à être utilisés dans le cadre desdits procédés, et l'utilisation desdites variantes à des fins thérapeutiques et diagnostiques.
