Vaccine conjugates are described comprising a capsular polysaccharide antigen conjugated to a carrier molecule, which carrier molecule is a protein associated with iron uptake by pathogenic microorganisms. The vaccine conjugate may comprise second or additional antigens also conjugated to the carrier. In a preferred embodiment, the carrier is a transferrin binding protein, in particular TbpA or TbpB. Also described are methods for conjugating the antigen/s to the carrier molecule, and to the use of transferrin binding protein in the manufacture of a carrier-antigen conjugate for vaccination. Affinity matrices comprising immobilised ligand for an iron uptake protein, and the use thereof for purifying a vaccine conjugate according to the present invention are also described.

L'invention concerne des conjugués de vaccins comprenant un antigène polysaccharide capsulaire conjugué à une molécule de transport, laquelle molécule est une protéine associée à une fixation de fer par des micro-organismes pathogènes. Le conjugué de vaccin peut renfermer un deuxième antigène ou des antigènes supplémentaires également conjugués avec la transporteuse. Dans un mode de réalisation préféré, la transporteuse est une protéine liant la transférine, en particulier TbpA ou TbpB. L'invention traite en outre de procédés permettant de conjuguer l'antigène, ou les antigènes, à la molécule de transport, et l'utilisation de la protéine liant la transférine dans la fabrication d'un conjugué d'antigène de transport destiné à la vaccination. Elle traite enfin de matrices d'affinité constituées de ligand immobilisé pour une protéine de fixation du fer, et de leur utilisation pour purifier un conjugué de vaccin.
