The present invention is directed to a new PTEN opener or a new opener of a large conductance Ca2+- activated K+ channel (Maxi-K channel) which comprises as an active ingredient a tetrazolylalkoxy-dihydrocarbostyril compound of the formula (I): (see formula (I) wherein R is cycloalkyl, A is lower alkylene, and the bond between 3- and 4-positions of the carbostyril nucleus is a single bond or a double bond, or a salt thereof. The PTEN opener is useful as a medicament for promotion of the survival of normal cells, brain cells, heart cells, and skin, and further for inhibiting of Gram negative sepsis and cell migration and cell invasion due to inhibition of PTEN. It is further useful as a medicament for the treatment of neuronal disorders, for example, an anticonvulsant, a neuroprotecting agent, a medicament for treatment of regional cerebral edema and neurologic motor impairment, cognitive disorders, traumatic brain injury, Parkinson's disease, epilepsy, migraine, and Alzheimer's disease, etc.
L'invention porte sur un nouvel agent régulateur de PTEN ou d'un canal K?+¿ activé par Ca?2+ ¿d'une grande conductibilité (canal maxi-K), ce nouvel agent régulateur comprenant comme ingrédient actif un composé de tétrazolylalcoxy-dihydrocarbostyrile de formule (I) dans laquelle R représente cycloalkyle, A représente alkylène inférieur et la liaison entre les positions 3- et 4- du noyau de carbostyrile est une liaison simple ou double, ou un sel de celui-ci, ce composé étant utile comme médicament destiné à favoriser la survie des cellules normales, des cellules du cerveau, des cellules du coeur et de la peau, et également à inhiber la sepsie Gram négatif et la migration cellulaire et l'invasion cellulaire imputable à l'inhibition de PTEN. Ledit composé est également utile comme médicament pour traiter les troubles neuronaux, par exemple, un anticonvulsivant, un agent neuroprotecteur, un médicament pour traiter l'oedème cérébral régional et la détérioration neurologique, les troubles cognitifs, les lésions cérébrales traumatiques, la maladie de Parkinson, l'épilepsie, la migraine et la maladie d'Alzheimer, etc.
