The present application relates to a process for purification of a bacterial cytolysin comprising the steps of a) growing a culture of cells expressing bacterial cytolysin, b) mechanically breaking the culture of cells to form an extract, c) prefiltering the extract, d) binding soluble aggregated bacterial cytolysin contained in the extract in the presence of detergent to a hydrophobic interaction chromatography material under high salt (preferably 0.6-2M salt) conditions, and e) eluting bacterial cytolysin in the presence of detergent under low salt (preferably 0-0.2M salt) conditions.

L'invention concerne un procédé pour purifier une cytolysine comprenant les étapes suivantes : a) croissance en culture de cellules exprimant une cytolysine bactérienne, b) interruption mécanique de la culture des cellules pour former un extrait, c) préfiltrage de l'extrait, d) liaisons de la cytolysine bactérienne agrégée soluble contenue dans l'extrait en présence de détergents à un matériau de chromatograhie d'interaction hydrophobe dans des conditions hautement salines (de préférence 0.6-2M de sel), et e) élution de la cytolysine bactérienne en présence d'un détergent dans des conditions faiblement salines (de préférence 0-0.2M de sel).
