A method for producing a refolded, inactive form of recombinantly produced NS2/3 protease which comprises the steps of: a) purifying the protease from inclusion bodies in the presence of a chaotropic agent; and b) refolding the purified protease by contacting it with a reducing agent and lauryldiethylamine oxide (LDAO) in the presence of reduced concentration of chaotropic agent or polar additive. The invention further comprises a method for activating this refolded inactive NS2/3 protease by adding an activation detergent. This method produces large amounts of the active NS2/3 protease to allow small molecules and ligands to be screened as potential inhibitors of NS2/3 protease, which may be useful as therapeutic agents against HCV.

L'invention concerne un procédé relatif à l'élaboration de NS2/3 protéase inactive repliée de recombinaison, qui comprend les étapes suivantes: a) purification de la protéase pour éliminer les corps d'inclusion en présence d'un agent chaotropique; et b) repliage de la protéase purifiée par contact avec un agent réducteur et de l'oxyde de lauryldiéthylamine en présence de concentration réduite d'agent chaotropique ou d'additif polaire. L'invention concerne également un procédé relatif à l'activation de cette NS2/3 protéase inactive repliée par adjonction d'un détergent d'activation. Le procédé décrit permet de fournir des quantités importantes de NS2/3 protéase active pour assurer le criblage des molécules et des ligands de petite taille en tant qu'inhibiteurs potentiels de NS2/3 protéase, lesquels peuvent être utiles comme agents thérapeutiques contre le VHC.
