The present invention relates to a method for purifying recombinant HCV single or specific oligomeric envelope proteins selected from the group consisting of E1 and/or E2 and/or E1/E2, characterized in that upon lysing the transformed host cells to isolate the recombinantly expressed protein a disulphide bond cleavage or reduction step is carried out with a disulphide bond cleavage agent. L'invention concerne un procédé de purification de protéines d'enveloppe oligomériques recombinantes du virus de l'hépatite C (VHC) simples ou spécifiques, sélectionnées dans le groupe constitué par E1 et/ou E2 et/ou E1/E2, caractérisées en ce que, lors de la lyse des cellules hôtes transformées pour isoler la protéine exprimée de manière recombinatoire, une étape de clivage ou de réduction de la liaison disulphide au moyen d'un agent de clivage de liaison disulphide est effectuée. L'invention concerne également une composition isolée selon le présent procédé, ainsi qu'à l'application diagnostique et thérapeutique de ces compositions. L'invention concerne en outre l'utilisation de la protéine E1 du VHC et des peptides dans le prognostic et la surveillance de l'efficacité clinique et/ou du résultat clinique du traitement du VHC.
