The present invention relates to pyrazolo-quinazolines, characterized by an ortho-substituted-arylamino, heterocy-clylamino- or C3-C7 cycloalkylamino residue at 8 position and an aryl, heterocyclyl or C3-C7 cycloalkyl as substituent of a carbox-amide at 3 position of the molecula framework. The compounds of this invention modulate the activity of protein kinases and are therefore useful in treating diseases caused by dysregulated protein kinase activity, in particular MPS1/TTK. The present inven-tion also provides methods for preparing these compounds, pharmaceutical compositions comprising these compounds, and meth-ods of treating diseases utilizing pharmaceutical compositions comprising these compounds.
Cette invention concerne les pyrazolo-quinazolines, caractérisées par un résidu cycloalkylamino en C3-C7, hétérocyclylamino ou arylamino substitué en ortho en position 8, et un aryle, hétérocyclyle ou cycloalkyle en C3-C7 en substituant dun carboxamide en position 3 de la structure moléculaire. Les composés de linvention modulent lactivité des protéines kinases et sont donc utilisés dans le traitement des maladies causées par un dérèglement de lactivité des protéines kinases, en particulier MPS1/TTK. Linvention concerne également des procédés de préparation de ces composés, des compositions pharmaceutiques comprenant ces composés et des méthodes de traitement des maladies utilisant les compositions pharmaceutiques contenant ces composés.
