The present invention relates to (pyrido/thieno)-[f]-oxazepine-5-one derivatives having the general Formula (I) wherein R1, R2 and R3 are independently H or (C1-4)alkyl; Ar represents a fused thiophene or pyridine ring optionally substituted with one or more substituents selected from (C1- 4)alkyl, (C1-4)alkyloxy, (C1-4)alkyloxy(C1-4)alkyl, CF3, halogen, nitro, cyano, NR4R5, NR4COR6, and CONR4R5; R4 and R5 are independently H or (C1-4)- alkyl; or R4 and R5 form together with the nitrogen atom to which they are bound a 5-or 6-membered saturated heterocyclic ring, optionally containing a further heteroatom selected from O, S or NR6; R6 is (C1-4)alkyl; A represents the residue of a 4-7 membered saturated heterocyclic ring, optionally containing an oxygen atom, the ring being optionally substituted with 1-3 substituents selected from (C1-4)alkyl, (C1-4)-alkyloxy, hydroxy, halogen and oxo; or a pharmaceutically acceptable salt thereof. The invention also relates to pharmaceutical compositions comprising said derivatives, and to the use of these (pyrido/thieno)-[f]-oxazepine-5-one derivatives in the treatment of neurological diseases and psychiatric disorders which are responsive to enhancement of synaptic responses mediated by AMPA receptors in the central nervous system.
L'invention se rapporte à des dérivés de (pyrido/thiéno)-[f]-oxazépine-5-one représentés par la formule I dans laquelle R?1¿, R?2¿ et R?3¿ sont indépendamment H ou (C¿1-4?)alkyle ; Ar représente un noyau de thiophène ou de pyridine fusionné facultativement substitué par un ou plusieurs substituants choisis parmi (C¿1-4?) alkyle, (C¿1-4?)alkyloxy, (C¿1-4?)alkyloxy(C¿1-4?)alkyle, CF¿3?, halogène, nitro, cyano, NR?4¿R?5¿, NR?4¿COR?6¿, et CONR?4¿R?5¿ ; R?4¿ et R?5¿ étant indépendamment H ou (C¿1-4?)-alkyle ; ou R?4¿ et R?5¿ formant ensemble avec l'atome de nitrogène auxquels ils sont liés un noyau hétérocyclique saturé à membres 5- ou 6- ; contenant éventuellement un autre hétéroatome choisi parmi O, S ou NR?6¿ ; R?6¿ est (C¿1-4?)alkyle ; A représente le résidu d'un noyau hétérocyclique saturé à membres 4-7, contenant, le cas échéant, un atome d'oxygène, le noyau étant facultativement substitué par des substituants 1-3 choisis dans (C¿1-4?)alkyle, (C¿1-4?)-alkoxy, hydroxy, halogène et oxo ; ou un sel de ce derniers pharmaceutiquement acceptable. Cette invention concerne aussi des compositions pharmaceutiques comprenant ledits dérivés, et l'utilisation de ces dérivés de (pyrido/thiéno)-[f]-oxazépine-5-one dans le traitement des maladies neurologiques et des troubles psychiatriques qui réagissent à la facilitation des réponses synaptiques induites par les récepteurs AMPA dans le système nerveux central.
