Disclosed is a pyrroloazepine derivative or a salt thereof. The pyrroloazepine derivative is represented by the following formula (I): (see fig. I) wherein the ring P represented by (see fig. II) means a pyrrole ring represented by the following structure: (see fig. III) or (see fig. IV) in which R1 represents alkyl, cycloalkyl or the like, and R2 represents H or alkyl; the dashed line indicates the presence or absence of a bond; and, when the bond is present, Z2 is not present/and Z1 represents H but, when the bond is absent, Z1 and Z2 are both H; Z1 represents H and Z2 represents a group OR3 in which R3 represents H, alkyl, aralkyl or the like; Z1 and Z2 both represent groups SR4 in which R4 represents alkyl, aralkyl or the like; or Z1 and Z2 are combined together to represent O, a group NOR5 in which R5 represents H, alkyl or the like, or a C2-C3 alkylenedithio; A represents alkylene, alkenylene or alkynylene; and Y represents a group (see fig. V) in which W represents CH, C= or N, m stands for 0 or 1, n stands for 1, 2 or 3, G represents O, S, C=O, sulfinyl, sulfonyl, alkylene, alkenylene, acetal or the like; E1 and E2 represent H or lower alkyl group; and D represents an aromatic hydrocarbon or am aromatic heterocyclic ring. The compound (I) has strong serotonin-2 receptor antagonistic action and low toxicity and less side effects, and is useful as a therapeutic for circulatory diseases such as ischemic heart diseases, cerebrovascular disturbances and peripheral circulatory disturbances.

On divulgue un dérivé de la pyrroloazépine ou un sel de cette dernière. Le dérivé de la pyrroloazépine est représenté par la formule suivante (I) : (voir fig. I) où le noyau P (voir fig. II) correspond à un noyau pyrrole représenté par la structure suivante : (voir fig. III) ou (voir fig. IV) où R1 est un alkyle, un cycloalkyle ou un groupe semblable, et R2 est un H ou un alkyle; la ligne tiretée indique la présence ou l'absence d'une liaison; et, lorsque la liaison est présente, Z2 est absent et Z1 représente un H, mais lorsque la liaison est absente, Z1 et Z2 sont tous deux un H; Z1 est un H et Z2 est un groupe OR3 dans lequel R3 est un H, un alkyle, un aralkyle ou un groupe semblable; Z1 et Z2 sont tous deux des groupes SR4 dans lesquels R4 représente un alkyle, un aralkyle ou un groupe semblable; ou Z1 et Z2 sont combinés ensemble pour donner un O, un groupe NOR5 dans lequel R5 est un H, un alkyle ou un groupe semblable, ou un alcénylènedithio en C2-C3; A est un alkylène, un alkénylène ou un alkynylène; et Y représente un groupe (voir fig. V) dans lequel W représente CH, C= ou N, m correspond à 0 ou 1, n correspond à 1, 2 ou 3, G représente O, S, C=O, un groupe sulfinyle, sulfonyle, alkylène, alkénylène, acétal ou un groupe semblable; E1 et E2 sont un H ou un groupe alkyle inférieur; et D est un hydrocarbure aromatique ou un noyau aromatique hétérocyclique. Le composé (I) possède une forte activité antagoniste à l'égard du récepteur de la sérotonine-2, une faible toxicité et moins d'effets secondaires et il est utile pour le traitement des affections circulatoires telles les cardiopathies ischémiques, les affections cérébro-vasculaires et les affections du système circulatoire périphérique.
