The invention provides a new method of determining the presence of one or more polymorphic sites in a target nucleic acid sequence. The method comprises determining changes in the melting profile of an amplified product where the amplified product was amplified with a primer pair where one of the primers is labeled, e.g., with a fluorescent label. The method employs a probe that is labeled with a label that interacts with the label on the primer, e.g., a quencher that quenches the fluorescent label on the primer, such that the signal is quenched when the probe and primer are in close proximity. A change in the melting profile of an amplified product in comparison to a control reflects the presence of the polymorphic site.
L'invention concerne un nouveau procédé qui permet de déterminer la présence d'une ou de plusieurs sites polymorphes dans une séquence d'acides nucléiques cible. Ce procédé consiste à déterminer des changements dans le profil de fusion d'un produit amplifié, ce dernier ayant été amplifié par une paire d'amorces dont une est marquée, par exemple, par une marqueur fluorescent. Le procédé de l'invention fait intervenir une sonde marquée par un marqueur qui interagit avec le marqueur présent sur l'amorce, par exemple, un désactiveur qui désactive le marqueur fluorescent présent sur l'amorce, de façon que le signal soit désactivé lorsque la sonde et l'amorce sont disposées à proximité étroite l'une de l'autre. Un changement dans le profil de fusion d'un produit amplifié par comparaison avec un produit témoin indique la présence du site polymorphe.
