The present invention relates e.g., to QM-7 or QT-6 cells comprising a heterologous mutant nicotinic .alpha.7 acetylcholine receptor and/or a nucleic acid encoding it, or a fragment or variant thereof. In a preferred embodiment, the mutant nicotinic .alpha.7 acetylcholine receptor subunit has a mutation in the M2 domain. QM-7 and QT-6 cells of the invention are useful for, e.g., assays such as high throughput assays that measure the influx of cations, such as Ca++ ions, into a cell. Such assays can be used, e.g., to identify agents that modulate the expression and/or activity of a mutant cell-surface- expressed channel receptor (e.g., the nicotinic .alpha.7 receptor), and which thus modulate, e.g., among other functions, processes involved in the central nervous system, such as learning and memory.
L'invention concerne, par ex., des cellules QM-7 ou QT-6 contenant un récepteur hétérologue mutant nicotinique à l'acétylcholine .alpha.7 et/ou un acide nucléique codant pour ce récepteur, ou bien un fragment ou une variante de celui-ci. Dans un mode de réalisation préféré, la sous-unité du récepteur mutant nicotinique à l'acétylcholine .alpha.7 a une mutation dans le domaine M2. Les cellules QM-7 et QT-6 de la présente invention servent, par ex., dans des tests tels que des tests à haut rendement qui permettent de mesurer la pénétration de cations tels que les ions Ca?++¿ dans une cellule. Des essais de ce type servent, par ex., à identifier des agents qui modulent l'expression et/ou l'activité d'un récepteur mutant à canal à expression de surface (par ex., le récepteur nicotinique .alpha.7) et qui modulent ainsi, entre autres fonctions, des processus du système nerveux central tels que l'apprentissage et la mémoire.
