The present invention relates to upgraded microarrays comprising of single stranded nucleotide probes coupled with biotin molecules that are stabilized on reflective or non-reflective surfaces of solid substrates. The invention also provides undemanding hybridization methods that avoid reverse transcription and fluorescent dye labelling procedures that are required for conventional microarray tests. A unique post-washing method and reagents thereof ensures the detainment of double stranded hybrids, and the removal of unhybridized probes and testing nucleotides, thereby augmenting the sensitivity and specificity of the disclosed microarray. The intensities of signals radiating from the retained hybrids are proportionally amplified by the reflective surface of substrates during microarray scanning processes. Quantifications of testing targets are determined by the signal intensities normalized to that of the reference probes.
