The invention concerns drug precursors with antimalarial effect, characterised in that it consists in quaternary bis-ammonium salts of general formula (I) wherein A and A', identical or different, are respectively either a group A¿1? and A'¿1? of formula (1) wherein n = 2 to 4; R'¿1? is hydrogen, C¿1?-C¿5? alkyl, optionally substituted by an aryl, a hydroxy, an alkoxy, wherein the alkyl comprises 1 to 5 C, or aryloxy and W is halogen or a nucleofuge group; or a group A¿2? which represents formyl-CHO, or acetyl-COCH¿3?. B and B', identical or different, represent respectively either the group B¿1? and B'¿1?, if A and A' respectively represent A¿1?, A'¿1?, B¿1?, B'¿1? representing a group R¿1? which has the same definition as R'¿1? above, but cannot be a hydrogen atom; or respectively the groups B¿2? and B'¿2?, if A and A' represent A¿2?, B¿2? or B'¿2? being the group R¿1? as defined above, or a group of formula (2) wherein -Ra is RS- or RCO-, wherein R is a C¿1?-C¿6? alkyl, substituted if required, a phenyl or benzyl, wherein the phenyl is substituted if required, the latter being optionally substituted; R¿2? is hydrogen, C¿1?-C¿5? alkyl, or a -CH¿2?-COO- (C1-C5)alkyl group; and R¿3? is hydrogen, C¿1?-C¿5? alkyl or alkenyl, substituted if required, a phosphate, an alkoxy wherein the alkyl is a C¿1?-C¿3? alkyl, or aryloxy; or an alkyl (or aryl) carbonyloxy; or R¿2? and R¿3? form together a cycle with 5 or 6 C; R and R¿3? can be bound to form a cycle. .alpha. represents: either a single bond when A and A' represent A¿1? and A'¿1?: or when A and A' represent A¿2?, and B¿2? and B'¿2? Represent (3) either, when A and A' are -CHO or -COCH¿3? and B¿2? and B'¿2? are R¿1?, a group of formula (4) or a group of formula (5) wherein (a) represents a bond towards Z and (b) a bond towards the nitrogen atom. Z is a C¿9?-C¿21? alkyl, if required with insertion of one or several bonds, and/or one or several heteroatoms O and/or S and/or several aromatic cycles, and the pharmaceutically acceptable salts of said compounds. Said precursors and cyclized thiazolium derivatives are useful as antiparasitic medicines in particular antimalarial and antibabesiosis.
L'invention concerne des précurseurs de drogues à effet antipaludique, caractérisés en ce qu'il s'agit de sels de bis-ammonium quaternaire de formule générale (I) dans laquelle A et A', identiques ou différents, sont soit, respectivement, un groupe A1 et A'1 de formule (1) où n = 2 à 4; R'1 est hydrogène, alkyle en C1 à C5, éventuellement substitué par un aryle, un hydroxy, un alkoxy, dans lequel l'alkyle comprend de 1 à 5 C, ou aryloxy et W est halogène ou un groupe nucléofuge, soit un groupe A2 qui représente un formyle -CHO, ou acétyl -COCH3. B et B', identiques ou différents, représentent: soit respectivement les groupes B1 et B'1, si A et A' représentent respectivement A1 et A'1 , B1 et B'1 représentant un groupe R1 qui présente la même définition que R'1 ci-dessus, mais ne peut pas être un atome d'hydrogène, soit respectivement les groupes B2 et B'2, si A et A' représentent A2, B2 ou B'2 étant le groupe R1 tel que défini ci-dessus, ou un groupement de formule (2) dans lequel -Ra est RS- ou RCO-, où R est un alkyle en C1 à C6, le cas échéant substitué, un phényle ou benzyle, dans lequel le phényle est le cas échéant substitué, ceux-ci étant éventuellement substitués R2 est hydrogène, alkyle en C1 à C5, ou un groupe -CH2-COO-alkyl(C1 à C5); et R3 est hydrogène, alkyle ou alkényle en C1 à C5, le cas échéant substitué, un phosphate, un alkoxy, dans lequel l'alkyle est en C1 à C3, ou aryloxy; ou un alkyl (ou aryl) carbonyloxy; ou R2 et R3 forment ensemble un cycle à 5 ou 6 C; R et R3 peuvent être reliés pour former un cycle. .alpha. représente: soit une simple liaison, lorsque A et A' représentent A1 et A'1: ou lorsque A et A' représentent A2, et B2 et B'2 Représentent: soit, lorsque A et A' sont CHO ou COCH3 et B2 et B'2 sont R1, un groupement de formule (4), ou un groupement de formule (5) dans lesquels (a) représente une liaison vers Z et (b) une liaison vers l'atome d'azote. Z est un alkyle en C9 à C21, le cas échéant avec insertion d'une ou de plusieurs liaisons multiples, et/ou d'un ou plusieurs hétéroatomes O et/ou S, et/ou d'un ou de plusieurs cycles aromatiques, et les sels pharmaceutiquement acceptables de ces composés. Applications de ces précurseurs et des dérivés cyclisés de thiazolium comme médicaments anti- parasitaires en particulier antipaludiques et antibabésioses.
