Texaphyrins are provided for use as radiation sensitizers. Advantageous properties of texaphyrins for use as a radiation sensitizer include: i) a low redox potential which allows radiation-induced hydrated electrons to flow to texaphyrin rather than neutralizing hydroxyl radicals, allowing hydroxyl radicals to cause cellular damage, ii) a relatively stable texaphyrin radical that reacts readily to covalently modify neighboring molecules causing further cellular damage, iii) intrinsic biolocalization, and iv) indifference to the presence or absence of O2. These properties allow texaphyrins to be particularly effective for treating the hypoxic areas of solid neoplasms. Methods of treatment for an individual having a neoplasm or atheroma include the use of a texaphyrin as a radiation sensitizer and as an agent for photodynamic tumor therapy, or the use of a texaphyrin for internal and for external ionizing radiation. Novel texaphyrins are provided.

L'invention concerne des téxaphyrines permettant la sensibilisation aux rayonnements. Les téxaphyrines utilisées comme agents de sensibilisation aux rayonnements présentent les propriétés avantageuses suivantes: I) un faible potentiel redox qui permet aux électrons hydratés induits par les rayonnements de se déplacer vers la téxaphyrine et non pas vers les radicaux hydroxyles neutralisants, ce qui permet aux radicaux hydroxyle d'altérer les cellules, ii) un radical de texaphyrine relativement stable réagissant rapidement pour modifier, par covalence, les molécules adjacentes, ce qui poursuit l'altération des cellules, iii) une biolocalisation intrinsèque, et iv) une indifférence à la présence ou l'absence de O¿2?. Ces propriétés permettent aux téxaphyrines d'être particulièrement efficaces dans le traitement des zones hypoxiques des néoplasmes solides. Les procédés de traitement de sujets présentant un néoplasme ou un athérome comprennent l'utilisation d'une téxaphyrine comme sensibilisateur de rayonnements et comme agent pour le traitement photodynamique de la tumeur, ou l'utilisation d'une téxaphyrine pour les rayonnements ionisants internes et externes. L'invention traite également de nouvelles téxaphyrines.
