A method for identifying host genes and encoded proteins for potential targets for therapeutic intervention employs a Gene Search Vector that is either lentivirus or MMLV-based, and can be used to inter- rogate an entire cell genome without prior knowledge of the genomic se-quence. This Random Homozygous Gene Perturbation (RUGP) technique is rapidly verifiable and is used to identify potential host targets for inter- vention for influenza, HIV and other viral infections. Using Thermal As-symetric Interlaced (TAIL) - PCR, the period for identification of promis-ing targets is reduced from months to weeks or less. Specific targets in-cluding PTCH1, Robo1 and Nedd4 are reviewed in detail.

La présente invention concerne une méthode d'identification de gènes hôtes et de protéines codées comme cibles potentielles en vue d'une intervention thérapeutique qui utilise un vecteur de recherche de gènes soit lentiviral soit MMLV et qui peut être utilisée pour interroger un génome cellulaire entier sans connaissance préalable de la séquence génomique. Cette technique de perturbation aléatoire des gènes homozygotes (RHGP) est rapidement vérifiable et est utilisée pour identifier des cibles hôtes potentielles en vue d'une intervention contre la grippe, le VIH et autres infections virales. En utilisant la PCR à asymétrique thermique entrelacée (TAIL-PCR), la période d'identification de cibles prometteuses est réduite de quelques mois à quelques semaines ou moins. L'invention examine en détail des cibles spécifiques, notamment PTCH1, Robo1 et Nedd4.
