A method for the rapid assessment of organ status, including organ damage following immunological or toxicological insult, in a subject based on the detection of one or more isoenzymes of glutathione S-transferase (GST) in diverse biological fluids comprises contacting a particle-labelled anti-GST antibody specific for said isoenzyme with a sample of a biological fluid suspected of containing said isoenzyme, said antibody having a sensitivity sufficient to detect at least at picomolar amount of said isoenzyme, and capturing the particle-labelled antibody-isoenzyme complex on an immobilised capture antibody to generate a visually detectable signal. The method permits the detection or determination of GST isoenzymes typically in a time of the order of 30 minutes from initial contact with immobilised anti-GST antiboby. The method is simple to use and thus can be used, for example, by patients at home to monitor their levels of a particular GST isoenzyme following a liver or renal transplant.
Procédé d'évaluation rapide, chez un sujet, de l'état d'un organe, notamment dans le cas de lésions dues à une agression immunologique ou toxicologique, par détection d'une ou plusieurs isoenzymes de glutathion-S-transférase (GST) dans différents liquides biologiques. Ce procédé consiste à mettre en contact, d'une part, un anticorps anti-GST marqué par particules et spécifique de cette isoenzyme, et d'autre part, un échantillon de liquide biologique dont on soupçonne qu'il renferme ladite isoenzyme, ledit anticorps ayant une sensibilité suffisante pour détecter au moins une quantité picomolaire de ladite isoenzyme, et à capturer le complexe anticorps-isoenzyme marqué par particules, sur un anticorps de capture immobilisé, afin de produire un signal visible. Ledit procédé permet habituellement la détection ou la détermination des isoenzymes de GST dans un délai de l'ordre de 30 minutes à partir du premier contact avec l'anticorps anti-GST immobilisé. Ledit procédé est facile à mettre en oeuvre et peut, de ce fait, être utilisé par les patients à leur domicile pour contrôler leur taux d'une isoenzyme de GST particulière à la suite d'une transplantation hépatique ou rénale.
