An autologous vaccine to tumor cells is produced by transducing the tumor cells with a herpes simplex virus amplicon containing the gene for a cytokine to provide transient expression of the cytokine by the cells. The tumor cells may transduced with the herpes simplex amplicons ex vivo or in vivo. Preferred cytokines used in the method of the invention include interleukin-2 and GM-CSF. The tumor cells may also be transduced with a combination of amplicons containing genes for two or more different cytokines, for example interleukin-2 and interleukin 12.
Production d'un vaccin autologue contre les cellules tumorales en transférant dans les cellules tumorales un amplicon du virus herpès simplex contenant le gène d'une cytokine permettant aux cellules d'exprimer temporairement la cytokine. Cette transduction d'amplicons du virus herpès simplex dans les cellules tumorales peut se faire ex vivo ou in vivo. Parmi les cytokines utilisées de préférence dans cette méthode figurent l'interleukine-2 et le GM-CSF. On peut également transférer dans les cellules tumorales une combinaison d'amplicons contenant des gènes de deux ou plusieurs cytokines différentes, par exemple l'interleukine-2 et l'interleukine-12.
