Kit for the detection of a nucleotide sequence of interest comprising at least one nucleotide probe marked with a tracer. The kit contains, in suitable containers, a reagent essentially comprising a linear backbone copolymer having lateral substituents, whose chain consists of a first type of repetitive unit and at least one other type of repetitive unit, in which at least one part of the units of the first type have a lateral substituent comprising a nucleotide unit, such a lateral substituent not being present on the other types of units. Each of said nucleotide units, all of which are identical, comprise at least one nucleotide sequence capable of hybridizing with said sequence of interest and nucleotide sequence capable of hybridizing with said probe, the reagent containing on average more than two of said nucleotide units, in molar equivalents, per mole of polymer. Such a reagent enables signal amplification to be obtained, and therefore lowers the sensitivity threshold. Application, in particular, in the production of tests for the detection of pathogenic organisms, or in the diagnosis of genetic diseases.
Nécessaire pour la détection d'une séquence nucléotidique d'intérêt, comprenant au moins une sonde nucléotidique marquée avec un traceur. Il contient, dans des conteneurs appropriés, un réactif comprenant essentiellement un copolymère à squelette linéaire portant des substituants latéraux, dont la chaîne est constituée par un premier type de motifs répétitifs et par au moins un autre type de motifs répétitifs, dans lequel au moins une partie des motifs du premier type portent un substituant latéral comprenant une unité nucléotidique, un tel substituant latéral étant absent sur les autres types de motifs, chacune desdites unités nucléotidiques, toutes identiques, comprenant, au moins, une séquence nucléotidique capable d'hybridation avec ladite séquence d'intérêt et une séquence nucléotidique capable d'hybridation avec ladite sonde, ledit réactif contenant en moyenne plus de deux desdites unités nucléotidiques, en équivalents molaires, par mole de polymère. Un tel réactif permet d'obtenir une amplification du signal, et donc d'abaisser le seuil de sensibilité. Application notamment dans la réalisation de tests de détection d'organismes pathogènes, ou de diagnostic de maladies génétiques.
