Disclosed are a method for measuring the amount of cholesterol in low density lipoproteins (LDL) in samples specifically and accurately and also a reagent used for the method. The method and the reagent employ cholesterol esterase and cholesterol oxidase, at least one of which is modified by binding to a sugar compound. By using such an enzyme, LDL-cholesterol can be measured directly using so far widely used automatic analyzer, which has been impossible in known measurement methods.
