This invention identifies tumor senescence genes induced by treatment with cytotoxic agents. The invention provides reagents and methods for identifying compounds that induce expression of these cellular genes and produce cellular senescence, particularly senescence in tumor cells. The invention also provides reagents that are recombinant mammalian cells containing recombinant expression constructs that express a reporter gene under the transcriptional control of a promoter for a gene the expression of which is modulated in senescent cells, and methods for using such cells to identify compounds that modulate expression of these cellular genes.
L'invention concerne des gènes de sénescence tumorale créés par traitement avec des agents cytotoxiques. La présente invention concerne plus particulièrement des réactifs et des procédés d'identification de composés provoquant l'expression de ces gènes cellulaires et produisant la sénescence cellulaire, notamment la sénescence dans des cellules tumorales. L'invention concerne par ailleurs des réactifs se présentant sous la forme de cellules mammifères de recombinaison, contenant des constructions d'expression de recombinaison exprimant un gène rapporteur sous le contrôle de transcription d'un promoteur d'un gène dont l'expression est modulée dans des cellules sénescentes. L'invention concerne également des procédés d'utilisation de telles cellules pour l'identification de composés modulant l'expression de ces gènes cellulaires.
