An adenovirus E1/E4 expressing packaging cell line is provided, which permits the generation of recombinant adenoviruses deleted in both gene regions. A method for enhancing the efficiency of transduction of a recombinant AAV into a target cell is provided by infecting a target cell with a recombinant AAV comprising a selected transgene under the control of regulatory sequences. The infected cell is contacted with an agent which facilitates the conversion of single stranded recombinant virus to its double stranded form.
Adénovirus E1/E4 exprimant une lignée cellulaire d'encapsidation, et permettant la production d'adénovirus de recombinaison à délétion dans les deux régions géniques. Le procédé visant à accroître l'efficacité de transduction d'un virus adéno-associé de recombinaison dans une cellule cible, selon la présente invention, consiste à infecter une cellule cible avec un virus adéno-associé de recombinaison comprenant un transgène choisi soumis à la régulation de séquences régulatrices. La cellule infectée est mise en contact avec un agent qui facilite la conversion d'un virus de recombinaison simple brin en une forme double brin du virus.
