Abstract The present invention relates to DNA compounds and expression vectors that encode deacetoxycephalosporin C synthetase (DAOCS) and deacetylcephalosporin C synthetase (DACS) activities. The compounds can be used to con- struct recombinant DNA expression vectors for a wide variety of host cells, including E. coli, Penicillium, and Cephalosporium. The invention also relates to the regulatory elements of the Cephalosporium acremonium DACS/DAOCS gene. A method for selecting Penicillium transformants, including those able to synthesize cephalosporin antibiotic due to the presence of the DACS/DAOCS-encoding DNA, is also provided. This transformation system utilizes an acetamidase gene for purposes of selecting transformants.
