The invention provides for the production of several humanized murine antibodies specific for the antigen Lewis Y, which is recognized by the murine antibody Lewis Y. The Lewis Y antigen is expressed in normal tissues but the level of expression is higher in certain tumour types so that the antigen can be used as a marker for cells of some breast, colon, gastric, esophageal, pancreatic, duodenal, lung, bladder and renal carcinomas and gastric and islet cell neuroendocrine tumours. The invention also provides for numerous polynucleotide encoding humanized Lewis Y specific antibodies, expression vectors for producing humanized Lewis Y specific antibodies, and host cells for the recombinant production of the humanized antibodies. The invention also provides methods for detecting cancerous cells (in vitro) and (in vivo) using humanized Lewis Y specific antibodies. Additionally, the invention provides methods of treating cancer using humanized Lewis Y specific antibodies.
L'invention concerne la production de plusieurs anticorps murins humanisés spécifiques contre l'antigène Lewis Y, lequel est reconnu par l'anticorps murin Lewis Y. L'antigène Lewis Y est exprimé dans des tissus normaux mais le niveau d'expression est plus élevé dans certains types de tumeurs, ce qui permet d'utiliser l'antigène comme marqueur de cellules pour certains cancers du sein, du côlon, de l'estomac, de l'oesophage, du pancréas, du duodénum, des poumons, de la vessie et du rein, et pour des tumeurs gastriques et des nésidioblastomes neuroendocriniens. L'invention porte également sur de nombreux polynucléotides codant les anticorps humanisés spécifiques contre Lewis Y, des vecteurs d'expression pour la production d'anticorps humanisés spécifiques contre Lewis Y, et des cellules hôtes pour la production par recombinaison desdits anticorps humanisés. Elle se rapporte également à des procédés de détection de cellules cancéreuses (in vitro) et (in vivo) au moyen des anticorps humanisés spécifiques contre Lewis Y, ainsi qu'à des méthodes de traitement du cancer au moyen de ces derniers.
