Two-domain MHC polypeptides useful for manipulation of antigen-specific T- cells are disclosed. These polypeptides include MHC class II-based molecules that comprise covalently linked .beta.1 and .alpha.1 domains, and MHC class I- based molecules that comprise covalently linked .alpha.1 and .alpha.2 domains. These polypeptides may also include covalently linked antigenic determinants, toxic moieties, and/or detectable labels. The disclosed polypeptides can be used to target antigen-specific T-cells, and are useful, among other things, to detect and purify antigen-specific T-celles, to induce or activate T-cells, and to treat conditions mediated by antigen-specific T-cells.
Cette invention se rapporte à des polypeptides MHC à deux domaines utiles pour la manipulation des lymphocytes T à spécificité antigénique. Ces polypeptides sont notamment des molécules basées sur le complexe d'histocompatibilité majeure (MHC) de classe 2 qui contiennent des domaines .beta.1 et .alpha.1 liés par covalence, et des molécules basées sur le complexe MHC de classe I qui comprennent les domaines .alpha.1 et .alpha.2 liés par covalence. Ces polypeptides peuvent également contenir des déterminants antigéniques liés par covalence, des fractions toxiques et/ou des étiquettes détectables. Ces polypeptides peuvent servir à cibler les lymphocytes T à spécificité antigénique et ils sont utiles notamment pour détecter et purifier les lymphocytes T à spécificité antigéniques, pour induire ou activer les lymphocytes T et pour traiter les états induits par les lymphocytes T à spécificité antigénique.
