A method of enhancing expression of a desired protein at mucosal effector sites, said method comprising placing the protein to be expressed under the control of a promoter having SEQ ID NO 2, SEQ ID NO 3 or SEQ ID NO 4 or a fragment or variant or any of these which has promoter activity, and causing expression in mucosal cells. Constructs used in the methods, as well as suitable recombinant gut-colonising microorganisms such as a Salmonella spp. are also described and claimed. Such organisms are useful in the preparation of vaccines.
L'invention porte sur un procédé visant à améliorer l'expression d'une protéine désirée au niveau des sites des muqueuses d'un effecteur. Ce procédé consiste à placer la protéine devant être exprimée sous la régulation d'un promoteur ayant les numéros de séquences: SEQ ID NO 2, SEQ ID NO 3 ou SEQ ID NO 4 ou un fragment ou une variante ou n'importe lequel de ceux-ci qui a l'activité du promoteur et à générer l'expression dans les cellules des muqueuses. L'invention porte également sur des produits de recombinaison utilisés dans ces procédés, ainsi que sur des micro-organismes de recombinaison appropriés colonisant les intestins tels que Salmonella spp., ces organismes étant utilisés dans la préparation de vaccins.
