Defective poxviruses that lack a function imparted by an essential region of its parental poxvirus can be employed for protein production and vaccination, among other uses. A polynucleotide encoding the protein to be produced by the recombinant virus is inserted into the defective poxvirus and placed under transcriptional control of a promoter. The defective poxvirus is viable when the lost function of the essential region is complemented by a host cell, transgenic animal or helper virus.
Selon l'invention, on peut utiliser dans la production de protéines et la vaccination, entre autres applications, des poxvirus défectifs auxquels il manque une fonction transmise par une région essentielle de leur poxvirus parental. Un polynucléotide codant la protéine qui doit être produite par le virus recombiné est inséré dans le poxvirus défectif et placé sous la commande transcriptionnelle d'un promoteur. Le poxvirus défectif est viable lorsque la fonction perdue de la région essentielle est complétée par une cellule hôte, un animal transgénique ou un virus auxiliaire.
