Disclosed is a method of expressing enzymatically-active, recombinant proteolytic tryptase in a eukaryotic host cell, expression constructs which drive the production of enzymatically-active tryptase in transformed hosts, and genetically-engineered eukaryotic host cells containing the expression constructs and which express enzymatically-active proteolytic tryptases. Uses for the proteolytic tryptases so produced are also disclosed. Also disclosed is a method of making active site mutants of proteolytic tryptases in a eukaryotic host cell, expression constructs which drive the production of the mutants in transformed eukaryotic host cells, and genetically-engineered eukaryoitic host cells containing the expression constructs and which express the active-site mutated form of proteolytic tryptases.
L'invention concerne un procédé permettant d'exprimer la tryptase protéolytique de recombinaison, enzymatiquement active dans une cellule hôte eucaryote, des structures d'expression qui pilotent la production de ladite tryptase dans des hôtes transformés, et des cellules hôtes eucaryotes mises au point génétiquement renfermant les structures d'expression, et qui expriment des tryptases protéolytiques, enzymatiquement actives. L'invention traite également des utilisations des tryptases ainsi produites. Elle concerne en outre un procédé permettant de fabriquer des mutants de site actif des tryptases protéolytiques dans une cellule hôte eucaryote, des structures d'expression qui pilotent la production des mutants dans des cellules eucaryotes transformées, et des cellules hôtes eucaryotes mises au point génétiquement contenant les structures d'expression et qui expriment la forme mutante de site actif des tryptases protéolytiques.
