The present invention provides recombinant replicable vesiculoviruses. The invention provides a method which, for the first time, successfully allows the production and recovery of replicable vesiculoviruses, as well as recombinant replicable vesiculoviruses, from cloned DNA, by a method comprising expression of the full-length positive-strand vesiculovirus antigenomic RNA in host cells. The recombinant vesiculoviruses do not cause serious pathology in humans, can be obtained in high titers, and have use as vaccines. The recombinant vesiculoviruses can also be inactivated for use as killed vaccines.
L'invention se rapporte à des vésiculovirus de recombinaison dont la réplication est possible. On décrit un procédé qui, pour la première fois, permet de produire et de recueillir des vésiculovirus à réplication possible, ainsi que les mêmes virus de recombinaison, à partir d'ADN cloné. Ce procédé consiste à exprimer dans les cellules hôtes l'ARN antigénomique à brin positif pleine longueur de ces vésiculovirus. Ces vésiculovirus de recombinaison, qui ne déclenchent aucune pathologie grave chez l'être humain, peuvent être obtenus avec une teneur élevée et sont utiles dans les vaccins. On peut aussi les inactiver afin de les utiliser comme vaccins tubés.
