A membrane-based assay device for detecting the presence or quantity of an analyte residing in a test sample is provided. The device utilizes a chromatographic zone on which is disposed a plurality of microporous particles. The chromatographic zone can effectively reduce the "hook effect" in a simple, efficient, and relatively inexpensive manner. In particular, the plurality of microporous particles allows larger-sized analyte/probe complexes to reach the detection zone before the uncomplexed analyte. Because the uncomplexed analyte is substantially inhibited from competing with the complexes for the binding sites at the detection zone, the incidence of "false negatives" may be limited, even at relatively high analyte concentrations.

L'invention concerne un dispositif d'essai, comprenant une membrane, destiné à détecter la présence ou la quantité d'un analyte résident dans un échantillon test. Ledit dispositif utilise la zone chromatographique sur laquelle est disposée une pluralité de particules microporeuses. La zone chromatographique peut effectivement réduire l'effet crochet d'une manière simple, efficace, et relativement peu coûteuse. En particulier, la pluralité de particules microporeuses permet à des complexes analyte/sonde de dimensions plus large d'atteindre la zone de détection avant l'analyte non complexé. Du fait que l'entrée en concurrence de l'analyte non complexé avec les complexes pour les sites de liaison sur la zone de détection soit sensiblement inhibée, l'incidence de faux négatifs peut être limitée, même dans des concentrations élevées d'analytes.
