An invention is described that permits the regulation of recombination in cells, organisms or appropriate cell-free systems. The invention involves creating fusion proteins between recombinase proteins, or components of recombinase systems, and ligand binding domains derived from nuclear receptors. The fusion proteins show little recombinase activity in the absence of the ligand that binds to the ligand binding domain. Upon binding of the ligand, recombinase activity is induced. The invention provides a practical means to regulate recobination in cells and organisms and, by linking ligand binding to recombination, provides a simple means whereby ligand binding can be measured as recombination achieved.
La présente invention permet de réguler une recombinaison effectuée dans des cellules, des organismes ou des systèmes acellulaires appropriés. Elle consiste à créer des protéines hybrides en fusionnant des protéines de recombinases, ou des éléments de systèmes de recombinases, avec des domaines de fixation de ligands dérivés de récepteurs nucléaires. Les protéines hybrides ont peu d'activité de recombinase en l'absence du ligand qui les fixe à son domaine de fixation. Elles sont amenées à cette activité de recombinase par la fixation de ce ligand. L'invention offre un moyen pratique de réguler une recombinaison effectuée dans une cellule ou un organisme; de plus, en liant la fixation d'un ligand à la recombinaison, elle constitue un moyen simple de mesurer la fixation d'un ligand en termes de réussite de la recombinaison.
