A composition for targeting one or more infectious agent(s) or target cell(s) comprising a complementary set of adenoviral vectors, wherein each vector is dependent upon the other for replication. The first vector of the pair may comprise an E1 expression cassette where E1 is expressed within one or more infectious agent(s) or target cell(s) from a promoter and does not express other adenoviral proteins. The second vector provides adenoviral proteins required for packaging and replication. As such, the composition is useful for the delivery and expression of prophylactic and/or therapeutic genes, some of which demonstrate direct cytotoxic effects, indirect cytotoxic effects, or sensitize target cells to the effects of other treatments. The composition may encode one or more antigen(s), with or without immunomodulatory genes, to induce one or more immune response(s) to the encoded antigen or antigens.
L'invention concerne une composition permettant de cibler un ou plusieurs agent(s) infectieux ou une ou plusieurs cellule(s) cible comprenant un ensemble complémentaire de vecteurs adénoviraux, chaque vecteur dépendant de l'autre pour se répliquer. Le premier vecteur de la paire peut comprendre une cassette d'expression E1, E1 étant exprimé dans un ou plusieurs agent(s) infectieux ou dans une ou plusieurs cellule(s) à partir d'un promoteur, et n'exprimant pas d'autres protéines adénovirales. Le second vecteur fournit les protéines adénovirales nécessaires pour l'encapsidation et la réplication. De ce fait, on utilise ladite composition pour distribuer et exprimer des gènes prophylactiques et/ou thérapeutiques, certains d'entre eux montrant des effets cytotoxiques directs ou indirects, ou sensibilisant des cellules cible aux effets d'autres traitements. Cette composition peut coder un ou plusieurs antigène(s) avec ou sans gènes immunomodulateurs afin d'induire une ou plusieurs réponse(s) immunitaires aux gènes ou antigènes codés.
