The present invention relates to replicons or self-replicating RNA molecules, derived from the genome of cardioviruses and aphtoviruses, which can be used to express heterologous proteins in animal cells. When injected in an animal host, for example in the form of naked RNA, these replicons permit the translation of the encoded heterologous protein. If the encoded heterologous protein is a foreign antigen, these replicons induce an immune response against the encoded heterologous protein. The invention uses cardiovirus and aphtovirus genomes to construct these replicons. The invention demonstrates that these replicons, when injected as naked RNA, can induce immune responses against a replicon-encoded heterologous protein in an animal recipient without the help of any kind of carrier or adjuvant.

L'invention concerne des molécules d'ARN autoréplicatives ou des réplicons dérivés du génome de cardiovirus et d'aphthovirus pouvant être utilisés pour exprimer des protéines hétérologues dans des cellules animales. Lorsqu'ils sont injectés dans un animal hôte, sous forme d'ARN nu par exemple, ces réplicons permettent la traduction de la protéine hétérologue codée. Si cette protéine hétérologue codée est un antigène étranger, ces réplicons induisent une réponse immunitaire contre la protéine hétérologue codée. L'invention concerne également l'utilisation de génomes de cardiovirus et d'aphthovirus pour construire ces réplicons. L'invention permet de démontrer que ces réplicons, lorsqu'ils sont injectés sous forme d'ARN nu, peuvent induire une réponse immunitaire contre une protéine hétérologue codée par un réplicon chez un animal receveur, sans l'aide d'aucun porteur ou adjuvant.
