ABSTRACT OF THE DISCLOSURE A restriction enzyme having a specificity such that the enzyme can recognize a palindromic hexanucleotide sequence 5'-GGCGCC-3' 3'-CCGCGG-5' in a double-stranded deoxyribonucleic acid, and can cleave the phosphodiester bonds contained in the double-stranded deoxy- ribonucleic acid between the two cytosines at the 3' terminal nucleotides indicated by arrows on the following sequence 5'-GGCGCC-3' 3'-CCGCGG-5' so that deoxyribonucleic acid fragments each having tetra- nucleotide long 3'-protruding terminal nucleotides (GCGC-3') are generated. The restriction enzyme is produced by culti- vating a strain belonging to the species Bifidobacterium breve, harvesting the resultant cells of the strain, separat- ing a crude extract from the cells, fractionating the crude extract, and purifying the fractionated crude extract for isolating therefrom the restriction enzyme. An example of the strain which can be used to produce the restriction enzyme according to the invention is B. breve named YIT4006 and deposited under Deposition No. FERM-P3906.
