The present invention relates to a composition for implantation in the subretinal space of an eye, the composition including amniotic membrane, which may be cryopreserved human amniotic membrane, and a plurality of retinal pigment epithelial (RPE) cells or RPE equivalent cells present at the amniotic membrane. The amniotic membrane may be intact, epithelially denuded, or otherwise treated. The invention includes the use of amniotic membrane for the culturing of RPE cells thereon, forming a surgical graft for replacement of Bruch's membrane as a substrate, and for the transplanting of RPE cells to the subretinal space. The composition does not elicit immunological reactions to alloantigens or to RPE specific autoantigens; and exerts anti-inflammatory, anti-angiogenic, and anti-scarring effects. The invention includes methods and kits for making or using composites including amniotic membrane and RPE cells. Also disclosed is a device for harvesting RPE cells.
La présente invention concerne une composition à implanter dans l'espace sous-rétinien d'un oeil. Cette composition contient une membrane amniotique, pouvant être une membrane amniotique humaine cryoconservée, et une pluralité de cellules épithéliales pigmentaires de la rétine (RPE) ou cellules RPE équivalentes présentes sur ladite membrane amniotique. Cette membrane amniotique peut être intacte, épithélialement dénudée ou traitée autrement. Cette invention concerne l'utilisation de la membrane amniotique pour la culture de cellules RPE présentes sur cette dernière, pour la formation d'un greffon chirurgical destiné à remplacer la membrane de Bruch comme substrat et pour la greffe de cellules RPE dans l'espace sous-rétinien. La présente composition ne suscite pas de réactions immunologiques face aux allo-antigènes ou auto-antigènes spécifiques des RPE et possède des effets anti-inflammatoires, anti-angiogéniques et anticicatrisants. Ladite invention concerne également des méthodes et des trousses pour la production ou l'utilisation de composites, contenant une membrane amniotique et des cellules RPE, ainsi qu'un dispositif de récolte de cellules RPE.
