The invention provides retroviral vectors that have enhanced transcription termination structures. The termination structures comprise one or several heterologous upstream transcription termination enhancer (UE) sequences, or one or more additional copies of endogenous UE sequences operably associated with the 3' LTR polyadenylation signal. The retroviral vectors of the invention have various improved properties over conventional vectors, including stronger gene expression, enhanced vector titer and reduced interference with host cell gene expression resulting from read-through of vector initiated transcriptional events.
L'invention concerne de nouveaux vecteurs r~troviraux pr~sentant des structures renforc~es de terminaison de transcription. Les structures de terminaison comportent au moins une s~quence UE activatrice h~t~rologue de terminaison de transcription en amont, ou au moins une copie additionnelle de s~quences UE endognes fonctionnellement associ~es au signal de polyad~nylation de longue r~p~tition terminale 3'. Ces vecteurs r~troviraux pr~sentent de meilleures propri~t~s par rapport aux vecteurs classiques, y compris une plus forte expression g~nique, un titre de vecteur renforc~ et des interf~rences r~duites avec l'expression g~nique de la cellule hÙte r~sultant de la lecture d'~v~nements transcriptionnels initi~s par les vecteurs.
