- 31 - ABSTRACT OF THE DISCLOSURE A bacterial strain contains and replicates therein a recombinant DNA plasmid comprising a vector and an RDS (Rhizobium diagnostic segment) derived from a symbiotic Sym plasmid recovered from a nitrogen fixing bacterial species. The disclosure also derives a recombinant DNA plasmid comprising the RDS and the RDS itself. The disclosure further describes a process for identifying the species of an unknown Rhizobium isolate by purifying the DNA of the isolate, cleaving the DNA of the isolate with a restriction endonuclease to generate isolate DNA restrict- ion fragments of various sizes, fractionating the isolate DNA restriction fragments on the basis of molecular size, incubating the isolate DNA restriction fragment fractions with a labelled DNA probe comprising the RDS of a known Rhizobium species, under hybridization conditions where substantially homologous DNA segments of at least 100 nucleotides length will hybridize to one another, and identifying the number of isolate DNA restriction fragments that hybridize with the RDS probe. In this way, the species of the Rhizobium isolate coincides with the Rhizo- bium species whose RDS probe hybridizes with more than one isolate DNA restriction fragment. The invention enables inexpensive and rapid method of screening bacterial iso- lates having the ability to nodulate one or more legume species.
