Methods of regulating the formation of heterochromatin at a target locus within the genome of a cell and methods of regulating the modification of a histone protein assumed on the DNA of a target locus within the genome of a cell are provided. In particular, the invention provides methods to regulate the structure of the genome of a cell, where the structure is regulated by modulating heterochromatin formation, heterochromatin architecture, histone methylation, and/or DNA methylation, by exposing the cell to an RNAi probe complementary to a target locus so as to activate the RNAi machinery to modify the genomic structure of the target locus and adjacent segments of the genome.
Cette invention concerne des méthodes de régulation de la formation de l'hétérochromatine dans un locus cible à l'intérieur du génome de la cellule ainsi que des méthodes de régulation d'une protéine histone présumée sur l'ADN dudit locus cible. L'invention concerne en particulier des méthodes de régulation de la structure par modulation de la formation de l'hétérochromatine, l'architecture de la structure de l'hétérochromatine, la méthylation de l'histone et/ou la méthylation de l'ADN, ceci par exposition de la cellule à une sonde ARN complémentaire d'un locus cible de manière à activer le mécanisme ARN et à modifier la structure génomique de ce locus et des segments adjacents du génome.
