Rolling circle replication of a padlock primer is inhibited when it is hybridised to a target nucleic acid that is long or circular. The invention provides methods of addressing this problem including cutting the target nucleic acid near or preferably at the site which hybridises with the padlock probe, whereby a 3'-end of the cut target nucleic acid acts as a primer for rolling circle replication of the padlock probe. Also included is a method for assaying for a polyepitopic target by the use of two affinity probes each carrying an oligonucleotide tag and of padlock probe for rolling circle replication in association with the two affinity probes.
Cette réplication en cercle roulant d'une amorce cadenas est empêchée lorsque cette amorce est hybridée à un acide nucléique cible qui est long ou circulaire. L'invention concerne des procédés visant à résoudre ce problème et consistant notamment à couper l'acide nucléique cible à proximité ou de préférence au niveau du site d'hybridation avec la sonde cadenas, de manière qu'une extrémité 3' de l'acide nucléique cible coupé agisse en tant qu'amorce de réplication en cercle roulant de la sonde cadenas. L'invention concerne encore un procédé de dosage d'une cible à plusieurs déterminants antigéniques, au moyen de deux sondes d'affinité portant chacune une étiquette oligonucléotidique et d'une sonde cadenas destinée à la réplication en cercle roulant, en association avec les deux sondes d'affinité.
