This invention relates to monovalent and multivalent, monospecific binding proteins and to multivalent, multispecific binding proteins. One embodiment of these binding proteins has one or more binding sites where each binding site binds with a target antigen or an epitope on a target antigen. Another embodiment of these binding proteins has two or more binding sites where each binding site has affinity towards different epitopes on a target antigen or has affinity towards either a target antigen or a hapten. The present invention further relates to recombinant vectors useful for the expression of these functional binding proteins in a host. More specifically, the present invention relates to the tumor-associated antigen binding protein designated RS7, and other EGP-1 binding-proteins. The invention further relates to humanized, human and chimeric RS7 antigen binding proteins, and the use of such binding proteins in diagnosis and therapy.

Cette invention se rapporte à des protéines de liaison monospécifiques monovalentes et multivalentes et à des protéines de liaison multispécifiques multivalentes. Un mode de réalisation de ces protéines de liaison comporte au moins un site de liaison, chaque site de liaison se liant à un antigène cible ou à un épitope sur un antigène cible. Un autre mode de réalisation de ces protéines de liaison comporte au moins deux sites de liaison, chaque site présentant une affinité, pour différents épitopes sur un antigène cible, mais aussi, soit pour un antigène cible, soit pour un haptène. La présente invention se rapporte en outre à des vecteurs recombinants convenant à l'expression de ces protéines de liaison fonctionnelle dans un hôte. La présente invention se rapporte plus particulièrement à une protéine de liaison d'antigène associé à la tumeur, désignée RS7, et d'autres protéines de liaison EGP-1. L'invention se rapporte enfin à des protéines se liant à l'antigène RS7, humanisés, humains et chimériques, et l'utilisation de telles protéines de liaison en diagnostic et en thérapie.
