The present invention relates to novel rumen bacterial mutants resulted from the disruption of a lactate dehydrogenase gene (ldhA) and a pyruvate formate- lyase gene (pfl) (which are involved in the production of lactic acid, formic acid and acetic acid) from rumen bacteria; a novel bacterial mutant (Mannheimia sp. LPK7) having disruptions of a lactate dehydrogenase gene (ldhA), a pyruvate formate-lyase gene (plf), a phosphotransacetylase gene (pta), and a acetate kinase gene (ackA); a novel bacterial mutant (Mannheimia sp. LPK4) having disruptions of a lactate dehydrogenase gene (ldhA), a pyruvate formate-lyase gene (pfl) and a phosphoenolpyruvate carboxylase gene (ppc) involved in the immobilization of CO2 in a metabolic pathway of producing succinic acid; and a method for producing succinic acid, which is characterized by the culture of the above mutants in anaerobic conditions. The inventive bacterial mutants have the property of producing succinic acid at high concentration while producing little or no organic acids, as compared to the prior wild-type strains of producing various organic acids. Thus, the inventive bacterial mutants are useful as strains for the industrial production of succinic acid.

L'invention concerne de nouveaux mutants bactériens de la panse produits par disruption d'un gène de lactate déshydrogénase (<i>ldhA</i>) et d'un gène de pyruvate formate-lyase (<i>pfl</i>) (qui sont impliqués dans la production d'acide lactique, d'acide formique et d'acide acétique) pour des bactéries de la panse; un nouveau mutant bactérien (<i>Mannheimia</i> sp LPK7) ayant les disruptions d'un gène de lactate déshydrogénase (<i>ldhA</i>), d'un gène de pyruvate formate-lyase (<i>pfl</i>), d'un gène de phosphotransacétylase (<i>pta</i>), et d'un gène d'acétate kinase (<i>adkA</i>); un nouveau mutant bactérien (<i>Mannheimia</i> sp LPK4) ayant les disruptions d'un gène de lactate déshydrogénase (<i>ldhA</i>), d'un gène de pyruvate formate-lyase (<i>pfl</i>) et d'un gène de carboxylase phosphoénolpyruvate (<i>ppc</i>) impliqué dans l'immobilisation de CO¿2? dans un trajet métabolique de production d'acide succinique. L'invention concerne également un procédé de production d'acide succinique, qui est caractérisé par la culture des mutants susmentionnés dans des conditions anaérobies. Les mutants bactériens de l'invention présentent des propriétés de production d'acide succinique à concentration élevée tout en produisant peu ou pas d'acide organique, par rapport aux souches de type sauvage antérieur de production de divers acides organiques. Ainsi, les mutants bactériens de l'invention sont utiles en tant que souches dans la production industrielle d'acide succinique.
