Abstract There are described ruthenium complexes to which an immunologically active material can be or is coupled. The ruthenium complex has the general formula Image I wherein L1, L2 and L3 are the same or different and each signify a bi- or polycyclic ligand with at least two nitrogen-containing heterocycles, whereby at least one of these ligands is substituted with at least one group conferring water-solubility, and whereby at least one of these ligands is substituted, optionally via a spacer group, with at least one reactive group, and whereby the ligands L1, L2 and L3 are attached to the ruthenium via nitrogen atoms. The ligands L1, L2 and L3 contain e.g. a 2,2'-bipyridine, 1,10-phenanthroline, benzbathophenan- throline or bathophenanthroline group. The spacer group is e.g. an alkylen group with a maximum of 8 C-atoms which is optionally substituted with -SO2-NH-, -S-, -O-, -COO- or -CO-NH-. The active group, to which the immunologically active material is coupled is e.g. a -COOH, -I, -NH2, -NCS or -SO2Hal group. As immunologically active material which can be coupled to the above mentioned ruthenium complexes come into consideration especially antigens, haptens or antibodies. e.g. antibodies to carcinoembryonic antigen. The ruthenium complexes according to the present invention can be detected with great sensitivity by fluorescence spectroscopy.
