Sarcodictyin A and B, eleutherobin, and bioactive analogs (A) thereof synthesized using solid phase and solution phase chemistries. The synthetic method employs an attachment of common precursors, e.g., compounds 1880 or 200, on a solid support for generating conjugates 230 and 240, followed by standard chemical manipulations. A combinatorial library of sarcodictyins and eleutherobin analogs is constructed with modified C-8 ester, C-15 ester and C- 4 ketal functionalities and is screened for activity with respect to tubulin polymerization and cytotoxic activity against tumor cells, including Taxol- resistant lines. Compounds 600, 610, 630, 660-700, 730, 760, 850, and 920 are identified to be of equal or superior biological activities as compared to their corresponding natural product.

L'invention concerne les sarcodictyines A et B, l'éleuthrérobine et des analogues (A) bioactifs de ces dernières, synthétisés par des procédés chimiques en phase solide et en phase de solution. Le procédé de synthèse comprend la liaison de précurseurs communs, p. ex. les composés 1880 ou 200, sur un support solide de manière à produire les conjugués 230 et 240, suivie de manipulations chimiques standard. On construit une banque combinatoire d'analogues de sarcodictyines et d'éleuthérobine à l'aide de fonctions ester C-8, ester C-15 et cétal C-4 modifiées et on crible cette banque en fonction de l'activité de polymérisation de tubuline et l'activité cytotoxique sur les cellules tumorales, y compris sur les lignées résistantes au Taxol. Les composés 600, 610, 630, 660-700, 730, 760, 850 et 920 sont identifiés comme présentant des activités biologiques égales ou supérieures à celles des produits naturels correspondants.
