There is disclosed assays to identify compounds that inhibit an enzymatic activity of MIF (macrophage migration inhibitory factor). The enzyme activity catalyzes a tautomerization of MIF-substrates, such as D-dopachrome to DHICA. In general, the assay is conducted in vitro by adding, mixing or combining MIF polypeptide and a suitable substrate in the presence or absence of a test compound, and measuring the tautomerization of the substrate. The test compounds that inhibit tautomerization in the assay are identified as MIF inhibitors.
L'invention concerne des méthodes de criblage pour l'identification d'une activité enzymatique du facteur d'inhibition de la migration des macrophages (MIF). L'activité enzymatique catalyse une tautomérisation de substrats porteurs du MIF, tels que le dopachrome, contre DHICA. En général, la méthode consiste à ajouter, mélanger ou combiner, in vitro, un polypeptide de MIF et un substrat approprié en présence ou en l'absence d'un composé d'essai, et à mesurer la tautomérisation du substrat. Les composés qui inhibent la tautomérisation dans le criblage sont identifiés en tant qu'inhibiteurs de MIF.
