The invention relates to a method for the rapid and simple identification of PUFA PKS in microorganisms. Said method is characterized by the fact that DNA sections representing PKS that produce specifically for PUFA (polyunsaturated fatty acids) are reproduced in vitro. The PUFA PKS-specific amino acid sequence LGIDSIKRVEIL makes it possible to derive oligonucleotides which are used for the efficient PCR screening for PUFA PKS genes or PUFA-producing microorganisms. The inventive method is particularly suitable for the high throughput screening of microorganisms for PUFA PKS genes.
L'invention concerne un procédé d'identification rapide et simple de PUFA-PKS dans des micro-organismes, consistant à multiplier in vitro des sections d'ADN spécifiques de PKS prdouisant PUFA (acides gras poly-insaturés). La séquence d'acides aminés LGIDSIKRVEIL spécifique à PUFA-PKS permet de dériver des oligonucléotides pour le criblage efficace par réaction en chaîne de la polymérase, visant l'obtention de gènes PUFA-PKS ou de micro-organismes produisant PUFA. Le procédé selon l'invention est particulièrement destiné à un criblage à débit élevé de micro-organismes, visant l'obtention de gènes PUFA-PKS.
