Methods for screening for substances capable of modulating the phosphorylation of tau protein are disclosed, and in particular paired helical filament (PHF) tau, and the use of such modulators in the treatment of tauopathies. The assays and screening methods are based on the identification of new phosphorylation sites in PHF tau and new kinases and combinations of kinases as therapeutic targets, in particular the identification of casein kinase 1 as a kinase which phosphorylates tau protein.

L'invention concerne des procédés de criblage de substances capables de moduler la phosphorylation de protéines tau, notamment de protéines tau à paires de filaments en hélice (PHF), et l'utilisation de tels modulateurs dans le traitement de tauopathies. Les procédés de dosage et de criblage reposent sur l'identification de nouveaux sites de phosphorylation dans des protéines tau PHF, de nouvelles kinases et de combinaisons de kinases en tant qu'agents thérapeutiques, notamment l'identification de caséine kinase 1 en tant que kinase phosphorylant les protéines tau.
