A method for preferentially observing an exposed position (1c) of a macromolecule. A sample is obtained having a macromolecule (1a) with a first proton (1) and a second molecule (2a) with a second proton (2); then applying a magnetic field (4) to the sample and irradiating the sample with a pulse sequence (5) that preferentially demagnetizes protons of the macromolecule (1, 3) relative to the second proton (2); allowing the second proton (2) to exchange (6) with an exposed proton (1) of the macromolecule; and detecting the magnetization from the relatively magnetized second proton (2), which is now bound to the exposed position (1c) of the macromolecule. The invention also provides a method for observing a position in the macromolecule that bind a ligand.

L'invention porte sur un procédé visant à observer la position exposée (1c) d'une macromolécule. Pour cela, on prélève un échantillon possédant une macromolécule (1a) pourvue d'un premier proton (1) et une seconde molécule (2a) pourvue d'un second proton (2). On applique ensuite un champ magnétique (4) sur l'échantillon et on irradie celui-ci d'une séquence d'impulsions (5) qui démagnétise de préférence des protons de la macromolécule (1, 3) par rapport au second proton (2), ce qui permet l'échange (6) du second proton (2) avec un proton exposé (1) de la macromolécule; et on détecte la magnétisation à partir du second proton (2) relativement magnétisé qui est ensuite lié à la position exposée (1c) de la macromolécule. L'invention porte également sur un procédé visant à observer une position de la macromolécule qui se lie à un ligand.
