Provided are means and methods for in vitro and in vivo detection of chemically induced effects on embryonic development and differentiation for the purpose of embryotoxicity/teratogenicity screening as well as for the identification of pharmaceuticals such as growth and tissue promoting factors based on differentiating pluripotent embryonic stem (ES) cells. The assays are based on the use of transgenic ES cells and non-human animals comprising such ES cells or derivatives thereof, wherein said ES cells are characterized by the differentiation-dependent expression of a secreted reporter protein; in particular secreted embryonic alkaline phosphatase (SEAP).
L'invention concerne des moyens et des procédés permettant de détecter in vitro et in vivo des effets induits chimiquement sur le développement et la différenciation embryonnaires, pour dépister une embryotoxicité/tératogénicité ainsi que pour identifier des substances pharmaceutiques telles que des facteurs favorisant la croissance et la formation de tissus, en fonction de la différenciation de cellules souches embryonnaires (ES) pluripotentes. Les tests sont fondés sur l'utilisation de cellules ES et d'animaux non humains transgéniques comprenant lesdites cellules ES ou des dérivés de celles-ci. Selon l'invention, les cellules ES sont caractérisées par l'expression, qui dépend de la différenciation, d'une protéine rapporteur sécrétée, en particulier d'une phosphatase alcaline embryonnaire sécrétée (SEAP).
