GLP-2 analogues are disclosed which comprise one of more substitutions as compared to h[Gly2]GLP-2 and which may have the property of an increased small intestine/colon and stomach/colon selectivity. More particularly, preferred GLP-2 analogues disclosed herein comprise substitutions at one or more of positions (11, 16, 20, 24) and/or (28) of the wild-type GLP-2 sequence, optionally in combination with further substitutions at position (2) and one or more of positions (3, 5, 7), and (10), and/or a deletion of one or more of amino acids (31) to (33) and/or the addition of a N-terminal or C-terminal stabilizing peptide sequence. The analogues are particularly useful for the prophylaxis or treatment of stomach and bowel-related disorders and for ameliorating side effects of chemotherapy.

La présente invention concerne des analogues du GLP-2 qui, comparés avec le h[Gly2]GLP-2, contiennent une ou plusieurs substitutions et sont susceptibles de présenter une sélectivité accrue vis-à-vis du petit intestin et du côlon, ainsi que de l'estomac et du côlon. Selon l'invention, les analogues préférés du GLP-2 contiennent plus particulièrement des substitutions au niveau de l'une ou de plusieurs des positions 11, 16, 20, 24 et/ou 28 de la séquence du GLP-2 de type sauvage, éventuellement combinées avec d'autres substitutions en position 2 et une ou plusieurs des positions 3, 5, 7, et 10, et/ou la délétion d'un ou de plusieurs des acides aminés 31 à 33 et/ou l'addition d'une séquence peptidique de stabilisation N-terminale ou C-terminale. Les analogues se révèlent particulièrement utiles pour la prophylaxie ou le traitement de troubles stomacaux et intestinaux, et pour améliorer les effets secondaires d'une chimiothérapie.
